GLP-2: a GLP-1 / GIP dual incretin reference

Why a single peptide that engages two incretin receptors changed body-composition research design.

Overview

GLP-2 is a 39-amino-acid synthetic peptide engineered as a dual agonist of the GLP-1 and GIP receptors. The combination is sometimes called a 'twincretin'.

Mechanism

GLP-1 receptor activation drives glucose-dependent insulin release, slows gastric emptying, and reduces appetite. Adding GIP receptor activity changes adipocyte signaling and energy expenditure, which is what distinguishes GLP-2 from single-receptor GLP-1 agonists in trial outcomes.

Notes for the lab

A fatty-acid sidechain anchors GLP-2 to serum albumin for an extended plasma half-life, enabling weekly dosing in human PK studies. Reconstitute in bacteriostatic water and use within ~30 days at 2–8 °C.
